Definition

CYP2C9 is responsible for the main metabolic pathway of S-warfarin; decreased-function alleles such as *2 and *3 reduce clearance and increase the risk of bleeding. Together, CYP2C9 and VKORC1 genotypes explain approximately 50% of the variation in warfarin dose; the dose can vary up to 10-fold between individuals.
