Definition

CYP1A2 is responsible for the oxidative metabolism of drugs such as caffeine, clozapine, olanzapine and theophylline. The main polymorphisms examined are variants such as CYP1A21C (-3860G>A, reduced inducibility) and CYP1A21F (-163C>A, increased inducibility with smoking); in addition, the TT genotype of -1545C>T (rs2470890) has been associated with significantly more severe side effects during clozapine treatment.
