Definition

The CERS1 gene encodes ceramide synthase 1, an enzyme that catalyzes C18-ceramide biosynthesis in the ER. Homozygous variants lead to a decrease in brain ceramide levels, causing EPM8, which presents in childhood to adolescence with action myoclonus, generalized tonic-clonic seizures, and slowly progressive cognitive decline.
