Definition

Immunodeficiency 74 (IMD74) is an X-linked immunological disorder resulting from hemizygous loss-of-function variants in the TLR7 gene. TLR7 is a Toll-like receptor that recognizes single-stranded viral RNA, including SARS-CoV-2; loss of function leads to an inadequate type I/II interferon response. Affected (usually young) males can develop severe respiratory failure in response to SARS-CoV-2 infection.
