Definition
Heterozygous pathogenic variants in the VCL gene can disrupt vinculin (and its heart-specific isoform, metavinculin), a cytoskeletal protein active in cell-cell junctions at costameres and intercalated discs, leading to dilated cardiomyopathy and familial hypertrophic cardiomyopathy. Vinculin plays a role in transmitting mechanical forces in cardiomyocytes.
