Definition

The CASP10 gene encodes the caspase-10 enzyme, which functions in Fas-mediated apoptosis; loss-of-function variants disrupt the cell death signal, leading to accumulation of autoreactive lymphocytes. The clinical picture includes lymphadenopathy, hepatosplenomegaly, and autoimmune cytopenias; there is an increased lifetime risk of Hodgkin and non-Hodgkin lymphoma.
