Definition

The SERPINE1 gene encodes plasminogen activator inhibitor-1 (PAI-1); the 4G/5G polymorphism in the promoter region determines PAI-1 expression levels. The 4G allele leads to higher PAI-1 transcription, increasing a thrombophilic tendency; the 4G/4G genotype has been associated with a risk of venous thromboembolism and coronary artery disease. It is not a strong predictor on its own.
