Definition

The VKORC1 gene encodes the vitamin K epoxide reductase complex, the direct target of warfarin. The -1639G>A (rs9923231) polymorphism in the promoter region reduces expression; because A allele carriers have low enzyme activity, they face a risk of excessive anticoagulation and bleeding at standard doses and require a lower initial warfarin dose.
