Definition

The SH2D1A gene encodes the adaptor protein SAP, which binds to SLAM family receptors on T and NK cells; when SAP is deficient, the inhibitory signals coming through these receptors are disrupted. X-linked lymphoproliferative disease type 1 is typically characterized by fulminant infectious mononucleosis triggered by EBV infection, hemophagocytic lymphohistiocytosis, dysgammaglobulinemia, and an increased risk of lymphoma.
