Definition

Biallelic pathogenic variants in the LPL gene cause deficiency of lipoprotein lipase, an enzyme found in capillary endothelium that hydrolyzes triglycerides in chylomicrons. Enzyme deficiency leads to severe chylomicronemia; the clinical picture includes recurrent abdominal pain, acute pancreatitis episodes, hepatosplenomegaly, and lipemia retinalis.
