Definition

The LIPE gene (19q13) encodes hormone sensitive lipase (HSL), responsible for hydrolysis of triglycerides and diacylglycerol in adipocytes. Biallelic loss of function variants cause familial partial lipodystrophy type 6 (FPLD6), which presents with variable fat excess in the face/neck/shoulders/upper trunk together with severe subcutaneous fat loss in the lower extremities. Adult onset progressive myopathy, type 2 diabetes, and hypertriglyceridemia may accompany it; this is a very rare form.
