Definition

The PLIN1 gene encodes perilipin-1, a protein that coats the surface of the lipid droplet in adipocytes. Frameshift mutations that produce a truncated, nonfunctional protein leave the lipid droplet abnormally unprotected and increase basal lipolysis, causing FPLD4; limb lipoatrophy, severe insulin resistance, and marked hypertriglyceridemia are seen.
