Definition

The PPARG gene encodes a major transcription factor governing adipocyte differentiation and lipid storage; it is also the target of thiazolidinedione class diabetes drugs. Dominant negative mutations impair adipocyte differentiation, causing FPLD3, with post-pubertal fat loss in the limbs, early and severe insulin resistance, and type 2 diabetes.
