Definition

Heterozygous pathogenic variants in the HNF4A gene impair the function of a transcription factor that regulates hepatic/pancreatic beta cell gene expression; MODY1 accounts for approximately 5% of all MODY cases. Some newborns develop transient hyperinsulinemic hypoglycemia, followed in young adulthood by diabetes due to progressive beta cell dysfunction; a good response to sulfonylureas is typical.
