Definition

The ELANE gene encodes neutrophil elastase; this serine protease plays an important role in neutrophil differentiation and granulopoiesis. Misfolded neutrophil elastase triggers ER stress, leading to apoptosis of promyelocytes; the result is severe chronic neutropenia and susceptibility to fatal pyogenic/fungal infections. ELANE mutations are the most common genetic cause of isolated severe congenital neutropenia.
