Definition

The CYP27A1 gene encodes the mitochondrial sterol 27-hydroxylase enzyme, responsible for converting cholesterol into cholic acid and chenodeoxycholic acid in the bile acid synthesis pathway. Biallelic loss-of-function variants reduce bile acid synthesis and lead to the accumulation of cholestanol in the brain, tendons and other tissues; the result is progressive neurological disorder, tendon xanthomas and early atherosclerosis.
